ABSTRACT

OBJECTIVE@#To study the effects of astragaloside-IV (As-IV) on the expression of inflammatory factor and proliferation of glomerular mesangial cells (GMCs) induced by angiotensin Ⅱ(AngⅡ).@*METHODS@#The model of diabetic nephropathy(DN) was mimic by angiotensin Ⅱ (10mol/L)inducing GMCs injury. Then the GMCs were treated with As-IV at different concentrations(25,50,100 μmol/L)for 48 hours. The proliferation of GMCs was detected by MTT. The level of reactive oxidative species (ROS) was measured by flow cytometry. The expression of monocyte chemoattractant protein-1(MCP-1) protein in supernatant was detected by enzyme- linked immunosorbent assay (ELISA). The expression of transforming growth factor-β1(TGF-β1) in GMCs was measured by Western blot.@*RESULTS@#Compared with model group, the proliferation of GMCs was inhibited in As-IV group. As-IV decreased the level of intercellular ROS, down-regulated the secretion of MCP-1 and the expression of TGF-β1 proteins.@*CONCLUSIONS@#As-IV could inhibit cell proliferation and inflammatory factors expression on GMCs induced by AngⅡ.

ABSTRACT

Objective: To explore the relationship between peripheral blood levels of Let-7e, miRNA-206, miRNA-605, miRNA-1236 and essential hypertension (EH) occurrence. Methods: Our research included in 2 groups: HE group, n=169 patients treated in our hospital from 2014-07 to 2016-12 and Control group, n=143 healthy subjects from physical examination at the same period. Blood pressure was measured by sphygmomanometer, the expression levels of peripheral Let-7e, miRNA-206, miRNA-605 and miRNA-1236 were detected by reverse transcription quantitative polymerase chain reaction (RT-qPCR). Relationship between miRNA levels and HE occurrence was studied by multivariate Logistic regression analysis. Results: Compared with Control group, EH group showed increased expressions of Let-7e and miRNA-605 (1.42±0.15) vs (1.00±0.21) and (2.14±0.22) vs (1.00±0.11), both P<0.05; decreased expressions of miRNA-206 and miRNA-1236 (0.69±0.12) vs (1.00±0.18) and (0.46±0.17) vs (1.00±0.31), both P<0.05. Correlation analysis presented that peripheral level of Let-7e was positively related to systolic blood pressure (r=0.562, P<0.05); miRNA-206 was negatively to systolic blood pressure (r=-0.582,P<0.05)and positively related to diastolic blood pressure(r=0.507,P<0.05);miRNA-605 was positively related to systolic blood pressure (r=0.625, P<0.05); miRNA-1236 was negatively related to diastolic blood pressure (r=-0.598, P<0.05) and positively related to BMI (r=0.690, P<0.05). Multivariate Logistic regression analysis indicated that elevated BMI could increase the risk of EH occurrence (OR=2.6412, 95%CI 1.7933-3.9941, P<0.01), elevated expression of Let-7e and miRNA-605 could increase the risk of EH occurrence (OR=1.6505 , 95%CI 1.5378-1.7632, P<0.05) and (OR=1.9893, 95%CI 1.5983-2.4203, P<0.01); reduced expression of miRNA-206 and miRNA-1236 could also increase the risk of EH occurrence (OR=0.7164, 95%CI 0.5546-0.8782, P<0.01)and(OR=0.4218,95%CI 0.2876-0.5614,P<0.01). Conclusion: Peripheral blood levels of Let-7e, miRNA-206, miRNA-605 and miRNA-1236 were closely related to EH occurrence.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the protective effect of Wufu Jingfang (WJ, containing Aconitum carmichaeli Debx, Radix Aconiti Lateralis Preparatae, Rhizoma Pinelliae, and snakegourd fruit) on myocardial ischemia-reperfusion injury (I/R) of rats, thus exploring the feasibility of recipes containing eighteen incompatible pairs for specific pathological conditions.</p><p><b>METHODS</b>Fifty male Wistar rats were randomly divided into five groups, i.e., the sham-operative control group (the SH group), the I/R group, the low dose WJ I/R group (the I/R +JFL group), the middle dose WJ I/R group (the I/R +JFM group), the high dose WJ I/R group (the I/R +JFH group), 10 in each group. Rats in the latter three groups were administered with WJ at 0.75 mL/100 g, 1.50 mL/100 g, and 3.00 mL/100 g body weight for 14 consecutive days by gastrogavage. All groups except the SH group received ligation of left anterior descending branch of coronary artery for 30-min ischemia followed by 120-min reperfusion. The micro-structural changes of myocardial mitochondria were observed by transmission electron microscope. The ischemic cardiomyocyte apoptosis was detected in each group using one-step terminal-deoxynucleotidyl transferase mediated nick end labeling (TUNEL). The mRNA expressions of B-cell leukemia/lymphoma 2 (Bcl-2) and Bcl-2 associated x protein (Bax) were detected by RT-PCR. The activities of lactic dehydrogenase (LDH) and creatine kinase (CK) were detected using ELISA. The myocardial infarct size was detected.</p><p><b>RESULTS</b>Compared with the I/R group, WJ pretreatment significantly suppressed the release of LDH and CK (Besides, the release of LDH and CK reduced along with increased dose.), reduced the myocardial infarct size, and lowered myocardial apoptosis index (P < 0.05). WJ pretreatment also modulated Bcl-2/Bax ratio by up-regulating Bcl-2 expression level while decreasing Bax expression level.</p><p><b>CONCLUSIONS</b>WJ pretreatment might protect the heart from I/R injury via decreasing myocardial cell apoptosis. The results suggested that eighteen incompatible pairs is not absolute, but relative. Chinese medical preparation containing opposite Chinese herbs could be used in specific pathological states such as ischemic cardiomyopathy.</p>
